Asante Kotoko came from behind to beat Dreams FC 3-1 in Obuasi on Saturday to go top of the Ghana Premier League table.
But a win for Medeama on Sunday against Liberty Professionals will see them back on top.
The Porcupine Warriors fell behind in the 38th minute as Agyenim Boateng scored after Dreams were awarded a controversial penalty.
A minute later, Fabio Gama, adjudged to have handled the ball in the box resulting in the penalty, redeemed himself with a defence-splitting pass to Emmanuel Gyamfi. The winger set up Brazilian striker Michael Vinicius to draw Kotoko level with a close-range header.
In the 56th minute, Kotoko benefited from a controversial decision as they were awarded a penalty. Defender Abdul Ganiu made no mistake from 12 yards.
Kotoko were in control of the game and increased their lead through Francis Andy Kumi in the 78th minute.
The defeat is the first suffered by Dreams since April 11 and they could drop out of the top four at the end of matchday 23.
